To describe the black guy as a 'homophobe' would be to prejudge the sexuality of the cross dresser. No?

Not really.
The actual sexuality of the 'blonde' doesn't matter.
It's the sexuality imposed on him by the transgressor that matters.
For example, people who are scared of clowns are also scared of people they think are clowns.

I think you do have a point.
There are plenty of people who are transphobic but not particularly homophobic.
Although we don't have any way of knowing whether he perceives them as trans, a cross-dresser, gay, a really jacked woman, a bad dresser, etc.
There was a case a while ago where some shitty kid lit an agender person in a skirt on fire on a bus in Oakland, CA. In interviews the kid kept saying stuff like "I don't like homosexuals..." and it was pretty clear he just perceived the victim as a gay boy. I think a lot of dicks like the guy in this video probably have that kind of unnuanced twattishness to them, and homophobe is an appropriate description. www.nytimes.com/2015/02/01/magazine/the-fire-on-the-57-bus-in-oakland.html?_r=0
