I don't think there's a formulaLooking back through "best of" QOTW answers I don't think there is a way to win. Some answers win because they are particularly heartwarming/poignant/apposite but are not meant to be entertaining and may not be particularly well-written (in an inventive use of language type of way). Others win with truly great humour, brilliantly written with laughs on every line. I've liked examples of all types.
I agree, in the humour stakes it does help if you're naturally funny, witty or have a raconteurial bent (ooer mrs, etc.) like Pooflake* and others. As to timing, one of my own front-pagers was posted late in the week but I'm sure the trend for winning posts is toward "the earlier the better".

*Or a colossal bell-end (like Pooflake).
